M-CSF Stress-Induced Priming of the Macrophage Cell

Stress is known to increase pro-inflammatory cytokine secretion. Inflammatory cytokines’ actions are down regulated by glucocorticoids. Colony stimulating factors, such as macrophage colony stimulating factor (M-CSF), are able to derive phenotypic changes in macrophage lineage cells based on the surrounding environment. In a chronic stress environment, previous studies have seen the macrophage phenotype to be CD11b+. The experiment used a social disruption stress (SDR) model in CD57BL-6 mice to induce the stress response and alter immune function as a consequence of stress. Cells were cultured with M-CSF. The CD11b+ phenotype of macrophages was confirmed via flow cytometry. CD11b+ were then stimulated with lipopolysaccharide (LPS). Cytokine levels of IL-6, IL-10, MCP-1, IFN-¿, TNF-a, and IL-12p70 were then measured using a cytometric bead array (CBA) assay. A student’s t-test was used to determine if cytokine production indicated any significant cytokine secretion indicating cell characteristics. The CBA assay showed no significant differences in the secretion of cytokines in stressed (SDR) mice versus home cage control (HCC) mice.
