Nucleosides play essential roles in DNA and RNA synthesis, metabolism, signal transduction, and immune modulation. Multiple regulatory proteins work together to sustain optimal nucleoside concentrations required to meet physiological demands. However, the contribution of solute carrier (SLC) proteins that transport hydrophilic nucleosides across lipid bilayers is often overlooked as key regulators of nucleoside action. The two major families of SLC proteins that translocate nucleosides across cell membranes are equilibrative nucleoside transporters (ENTs) and concentrative nucleoside transporters (CNTs). Recently, variation in the transporter function and expression of these nucleoside transporters (NTs) were associated with defects in blood cell formation and drug disposition. Here, we systematically identified ENTs and CNTs that are essential for the successful differentiation and production of functional blood cells, and we highlight a critical role for nucleoside transporters in nucleoside analog disposition through the generation and characterization of novel transporter-deficient murine models. Results from these studies support the clinical evaluation of NTs mutational status and expression in patients with blood disorders, provide a rationale for stratifying patients receiving nucleoside drugs, and underscore novel treatment strategies that modulate NT expression to improve drug efficacy.
