Abstract
Host-Rotaxanes function exceptionally well as cellular transport agents. Transport is mainly attributed to Host-rotaxane architecture, which consists of cyclophane pocket, DB24C8 wheel, and axle. The wheel needs to freely move along the axle. To function as a therapeutic device, however, a method to target cells is necessary. We propose that attaching a switch onto the wheel through an imine bond will control the motion of the wheel and enable the transporter to be turned on in acidic environments and turned off in basic environments. Human Body consists of wide range of pH values, which gives us a simple method to target various tissues or cell types. The first chapter describes the synthesis and characterization of Host-rotaxanes possessing an imine functionality. The second chapter will mainly deal with work involved in synthesis of novel Coumarin containing Host-rotaxanes. Coumarin derivatized rotaxanes will be used to determine the intracellular location of the rotaxane in the cells. The location can be monitored easily by observing coumarin’s fluorescence in the cells and can also be easily quantified by using methods such as flow cytometry. The future scope of the thesis and its development are discussed and recommendations are made.
