Abstract
The critical role of thyroid hormone (TH) in vertebrate development has been well shown in amphibian metamorphosis. When TH level increases, TH target tissues and organs undergo drastic developmental changes that are time- and tissue-specific. However, it has not been fully understood how the tissues can determine the timing and rate of developmental changes during metamorphosis. In this dissertation, I investigate the role of TH signaling proteins and TH receptors that have been hypothesized to regulate developmental timing and tissue-responsivity in TH-dependent development by examining Xenopus. First, I show the role of TH signaling proteins (TH transporters and cytoplasmic TH binding proteins) in the regulation of the timing of TH-dependent development. I examined various tissue-specific expression patterns and relative expression levels of the signaling proteins at different developmental stages. Using in-vivo functional analysis, I examined the role of the over-expressed signaling proteins in the timing of tissue transformation. Second, I show the role of unliganded TH receptors in developmental regulation. Using TRa knockout Xenopus tropicalis, I show that unliganded TRs are critical to the delaying of developmental initiation through the repression of TH-response genes. Altered levels of tissue responsivity to TH in TRa knockout animals show that unliganded TRs also contribute to the rate of tissue transformation in the early developmental period. Third, I dissect the role of TRa in development and growth rates during metamorphosis and show its impact on the determination of developmental completion. By examining intestinal development in TRa knockout animals, I show the tissue-specific role of TRa in TH-dependent development.
